چکیده

Maturational changes at the CD4 2 CD8 2 double negative (DN) to CD4 1 CD8 1 double positive (DP) transition are dependent on signals generated via the pre–T cell receptor (TCR) and the nonreceptor protein tyrosine kinase p56 lck (Lck). How Lck activities are stimulated or relayed after pre-TCR formation remains obscure. Our structure–function mapping of Lck thymopoietic properties reveals that the noncatalytic domains of Lck are specialized to signal efficient cellular expansion at DN to DP transition. Moreover, although substitution of the Lck catalytic domain with FynT sequences minimally impacts DP development, single positive thymocytes are most efficiently produced in the presence of kinases containing both the NH 2 -terminal and catalytic regions of Lck. These findings demonstrate that the Lck structure is uniquely adapted to mediate signals at both major transitions in thymopoiesis.

Lck Domains Differentially Contribute to Pre–T Cell Receptor (Tcr)–And TCR-α/β–Regulated Developmental Transitions

Maturational changes at the CD4(-)CD8(-) double negative (DN) to CD4(+)CD8(+) double positive (DP) transition are dependent on signals generated via the pre-T cell receptor (TCR) and the nonreceptor protein tyrosine kinase p56(lck) (Lck). Evaluation of Specific Purified TCR Effect on the Immunoregulatory Potential of TGF-beta

Transforming growth factor beta (TGF-b) is a mediator released by nearly all cell types. It has suppression activity on the immune system, but exactly how this effect is carried out is not clear. Previous experiments showed that IgG interacts with or carriers active TGF-b, that could suppresses cytotoxic T-cell responses to an immunogenic tumor in mice. Since T cell receptor (TCR) has structura...

Regulation of T cell development by c-Cbl: essential role of Lck.

A canonical pre-TCR/TCR signaling pathway critical for thymic T cell development involves sequential phosphorylation and signaling through Lck, Zap70, Lat and Slp76. However, we and others have previously reported that genomic deletion of c-Cbl (Cbl) partially or completely reverses the defects in thymic development in mice deficient in Zap70, Slp76, Lat or Vav1, indicating the presence of alte...

Essential role of the T cell–specific adapter protein in the activation of LCK in peripheral T cells

T cell-specific adapter protein (TSAd) is a SRC-homology-2 (SH2) domain-containing intracellular signaling molecule that is required for T cell antigen receptor (TCR)-induced cytokine synthesis in T cells. How TSAd functions in TCR signal transduction is not clear. Previous work has suggested a nuclear role for this adapter. However, other evidence suggests that TSAd also functions in the cytop...

Requirement of CD3 Complex–associated Signaling Functions for Expression of Rearranged T Cell Receptor β VDJ Genes in Early Thymic Development

During alpha beta thymocyte development, the clonotypic alpha beta-T cell receptor (TCR) is preceded by sequentially expressed immature versions of the TCR-CD3 complex: the pre-TCR, containing a clonotypic TCR-beta chain and invariant pre-Talpha, is expressed on pre-T cells before rearrangement of the TCR-alpha locus. Moreover, clonotype-independent CD3 complexes (CIC) appear on pro-T cells bef...
